1 Cleavage Test on Syntaxin 1a & SNAP-25

Three independent laboratory tests confirm CG-PTx’s enzymatic activity on key SNARE proteins. In human bone marrow neuroblastoma cells (SH-sy5y), western blot analysis demonstrated dose-dependent cleavage of both Syntaxin 1a and SNAP-25 at concentrations of 10nM, 50nM, and 100nM over 4 hours. A cell-free system test further confirmed cleavage of recombinant Syntaxin 1a. Immunocytochemistry at 100nM showed a significant reduction in the Syntaxin 1a green signal compared to control, visually confirming protein cleavage within neuronal cells.

Confocal fluorescence microscopy at x400 and x1400 magnification confirms that CG-PTx penetrates into both the cytosol and nucleus of neuronal cells. Using human bone marrow neuroblastoma cells (SH-sy5y) treated for 1 hour in serum-free DMEM, the rhodamine-labeled CG-PTx (red) is clearly visible inside the cells, merging with the DAPI nuclear stain (blue) to produce a pink signal — confirming intracellular delivery.
